Chapter 13 (pg 337-339) Transcription in Eukaryotes Eukaryotic RNA Polymerases: each RNA Pol recognizes different class of promoters RNA Pol I: exclusively in nucleolus; ctalyzes synth of some rRNA RNA Pol II: only in nucleoplasm; synthesizes mRNA and some snRNA RNA Pol III: only in nucleoplasm; synth tRNA, 1 rRNA & rest of snRNA Traits: multiple subunits encoded by several genes Transcription of Protein-Coding Genes by RNA Pol II (produce precursor mRNAs) Promoters and Enhancers Analysis of Promoters of Protein-Encoding gnees - examine effects of mut that delete/alter bp upstream from starting point of transcription to see if mut affects transcription - compare DNA seq upstream of several protein-coding genes to see if there are any consensus seq - results: 2 regions core promoter: cis-acting seq elem needed for trnscrp machinery to start RNA synth at correct site; w/in 50 bp upstream; w/ nothing else, low trnscrp levels; ex: Inr (initiator): spans transcription initiation site
