Gram Staining Materials: Crystal Violet, Grams Iodine, 95% Ethyl Alcohol, Safranin, Glass Slides, Bunsen Burner, Inoculating Loop, Lens Paper, Immersion Oil, Xylene Bacterial Cultures: Bacillus Thuringiensis G+ (always purple), Escherichia Coli G-, Serratia Marcescens, S. Aureus Procedure 1. Prepare 2 slides with two bacterial smears. 2. Cover smears for 1 minute with Crystal Violet 3. Rinse slides for 5 seconds, shake off excess water 4. Cover smear with gram’s iodine for 1 minute 5. Rinse again with water for 5 seconds 6. Decolorize smear with 95% ethanol by adding 1 drop at a time over sink (time 15-30 seconds) 7. Stop decolorizing and rinse slides with tap water 8. Counterstain each smear with safranin for 30 seconds 9. Rinse slides with tap water and blot dry with a paper towel Gram positive: violet Gram negative: red Materials: Glass slides, Carbolfuchsin, Acid alcohol, Methylene Blue, Clothespin, Paper towel, Hot plate, beaker Bacterial Cultures: Mycobacterium smegmatis, E. Coli
