James Huang February 9, 2010 6-9 Tue BME 2.204 Hall Locker: 295, Incubator Space: 301 Lab 1: Light Microscopy Microbiology Lab Questions Questions: 1) In my own experiences and observations I observed that there was agreement between the aspects of the table and what we observe in real life (during the experiment). I observed that when using the low power objective (10X) there was a relatively larger field of width than when in High or Oil immersion. When examining the Magnification at low power, then high dry, and then oil immersion I observed a decreasing width of field. Also, I would notice that certain objects could be “zoomed” in on by rotating the lens in that same order. The implication of this is that the magnification was increasing, like in the table. The focal lengths also must have been becoming increasingly short because the rays of light would have to be bent at more and more radical angles in order to still observe the image. This effect could be observed by noticing the decreasing working distance, when focusing on the
