The purpose of the Newcombe experiment is to determine if E. coli’s resistance to T2 is spontaneous or if the resistance occurs due to the bacteria’s exposure to the T2 bacteriophage. This helps us determine whether mutations in a population happen due to the exposure to an agent or are already present in some individuals, and therefore are being passed on to future generations. If the mutations occurred prior to exposure to T2 as hypothesized, then it would be expected that the plate that was respread with E.coli before being sprayed with T2 to have more colonies than the one that was not respread because respreading will disrupt the colonies and make them spread throughout the plate more.
