( For the lab practical exam, be sure you know how to properly perform each technique). Section 6.1: Isolation of Chloroplasts a. Prepare a Miracloth filter. b. Weigh 20 g of torn leaf blade tissue. c. Use a pre-chilled blender to produce a green slurry of spinach leaves. d. Filter the slurry through cheesecloth and the Miracloth filter. e. Follow proper cleaning procedures for the beaker, funnel, and blender to immediately remove green residue so that cell residue does not permanently stick to the glass. f. Centrifuge at RCF of 200 x g. g. Balance the tubes with the chloroplast supernatant on the two-pan balance. h. Centrifuge at RCF of 1100 x g to pellet Class II chloroplasts. i. Follow proper cleaning procedures for the centrifuge tubes to remove green residue. j. Use a tissue grinder and Teflon® pestle to homogenize the green liquid suspension. k. Prepare a wet mount slide of the chloroplast preparation. Section 6.2:
