( For the lab practical exam, be sure you know how to properly perform each technique). Section 9.1: Loading Protein samples into wells of a Polyacrylamide Gel a. Heat protein samples to 60 o C in a water bath. b. Know how to properly operate a microcentrifuge. c. Check wells of the gel to ensure they are in good condition. d. Attach the gel sandwich to the electrophoresis apparatus. e. Know how to forcefully wash the wells of the gel before loading samples. f. Know how to load samples directly into the wells of a gel without spillover into other lanes. g. Fill wells with Tank Buffer (electrophoresis running buffer). h. Move the gel apparatus to the power supply before turning on the power supply. i. Load each lane of the gel with 60 uL of the appropriate sample. j. Place the cover on the electrophoresis apparatus. k. Know how to attach electrode cables to the power supply. Section 9.2: Electrophoretic Separation of Proteins a. Multiply the desired milliAMPs by the number of gels that will be attached to the power supply. b.
