( For the lab practical exam, be sure you know how to properly perform each technique). Section 10.1.: Primary Antibody Binding to the Blocked Membrane a. Discard the primary antibody solution into a waste beaker. b. Wash the nitrocellulose membrane to remove unbound primary antibodies. c. Each wash cycle consists of steps 1 through 3 below: 1. Add 10 mL of Tris-buffered Saline (TBS) wash solution to the dish that contains the nitrocellulose filter. 2. Return the membrane to the rotary shaker for 5 minutes. 3. Discard the TBS wash solution. 4. Repeat steps 1-3 five times. Section 10.2.: Secondary Antibody Binding to the Primary Antibody. a. Add the Secondary Antibody Solution to the washed nitrocellulose and incubate on the shaker for 1.5 hours. b. Wash the nitrocellulose, as was done in Section 10.1, to remove unbound secondary antibodies.
