Bio Med October 27, 2008 Lab Write Up Title - Bacteria in Food: Bacterial Titer Determination Procedures: In order to make sure that the results come out accurately, you must do the correct serial dilution. Here is a step by step procedure for completing the refrigerated serial dilution. We want the plating concentrations to be 10^-3 and 10^-5. You will need three empty vials and two empty Petri dishes to do this dilution properly. Start with 10ml of the stock solution. In this case, your stock solution will be the meat that was given to you. This will be 10^-1. Add 9ml of nutrient broth to the first vial. Then, using a micropipette, transfer 1000ul into that first vial. This new vial is 10^-2 and is labeled 9ml. Then from your new vial, transfer 1000ul into a Petri dish, and label the dish 10^-3. Now, from your 10^-2 solution, transfer 100ul into a new vial. This new vial will have 9.9ml of nutrient broth in it. This vial is 10^-4 and is labeled 9.9ml. From this new vial, transfer 1000ul into another Petri dish and label this one 10^-5. That is the serial dilution for the refrigerated meat. The serial dilution for the Non-refrigerated meat isn’t that much different. You will need four empty vials and two empty Petri dishes.
