In flask 2, add 40g of dextrose (Sigma; St. Louis, MO) into 1L of dH 2 O. In flask 3, add at least 1.5L of dH 2 O. Loosely cap all three flasks 3) at the same time, autoclave the flasks at 121 o C for 25 minutes 4) after autoclaving, allow the flasks to cool to approximately room temperature, add the contents of flask two (containing dextrose) to flask one (containing tryptic soy broth), then q.s flask one up to 4L with flask three, containing dH 2 O. This results in tryptic soy broth with 1% (wt/vol) dextrose Tryptic soy broth with 1% dextrose 1. In a 1000mL flask - add: - 500mL of dH 2 O ……………………. Brain Heart Infusion broth (BHI) 1. In a 1000mL flask - add: - 500mL of dH 2 O ……………………. - LB broth 1. In a 1000mL flask - add: - 500mL of dH 2 O ……………………. Tryptic soy agar (TSA)
