3. Suppose you have a recombinant plasmid that you suspect might contain Exon 2 of the β -globin gene inserted into a vector as an EcoRI fragment. Also suppose you have a linear DNA fragment that is the authentic Exon 2 of the β -globin gene. Assume you know the entire genomic sequence of this gene. In part a) describe how you could use Southern blotting to test if the recombinant plasmid does have the Exon 2 insert. Give enough details to show that you understand the method. In part b) describe how you could use PCR to test if the recombinant plasmid does have the Exon 2 insert. For both parts be sure to describe the result expected if the Exon 2 insert is present in the plasmid, and if it isn’t. (20 points total) a) Southern blotting b) PCR 2
