Bioc100B Winter 2010 Rubin Problem Set #3 Due Tuesday, January 26 th 10:00 A.M. 1) Explain how each of the following observations is either consistent or inconsistent with the symmetry and sequential models of allosterism in ligand binding. a) In almost all crystal structures of hemoglobin (including both liganded and unliganded), all four subunits are in the same conformation. The idea that subunits in oligomeric proteins are all in the same conformation is consistent with one of the assumptions of the symmetry model. While one would not necessarily expect all of the subunits to be always in the same conformation in the sequential model, it could be the case (e.g. when all subunits are unbound or bound with ligand). b) Mutations that disrupt interactions between subunits often disrupt allosteric effects. In multisubunit proteins, allosteric effects arise from structural changes that propagate from subunit to subunit.
