Materials and Method DNA extraction is done by collecting buccal cells inside each student’s mouth. The students should avoid brushing teeth, eating or anything that involves the mouth for one hour prior to the laboratory so that the loose cell is not washed away and to obtain a maximum amount of cheek cell possible. The cheek cells are then added with the cell lysis solution to lyse the cell by fragmenting the membrane of the cells. The addition of Proteinase K in the tube after 15 minutes is to isolate the Deoxyribonucleic Acid (DNA) and Ribonucleic Acid (RNA) of the cell and inactivate the Deoxyribonucleases (DNases) and Ribonucleases (RNases). To remove the proteins in the cell lysate solution. Protein Precipitation Solution is added. The salt in the protein precipitation solution that contains a neutral salt induce the proteins to precipitate, producing a tight, greenish pellet after being centrifuged. Next the DNA precipitation is done by adding isopropanol and glycogen solution in a new tube with the supernatant from the previous tube in order to precipitate the DNA and ethanol is later added after centrifuging the new tube to wash away the salt from the protein precipitation solution. Centrifuging is done to collect the DNA sample at the bottom of the tube and the ethanol is poured off carefully to make sure the DNA pellet still stays inside the tube. Then the DNA Hydration Solution which contains pH 7.4 Tris Buffer and EDTA is added to the tube containg the DNA pellet to reduce DNA degradation and stabilize the DNA. After purifying the DNA from the buccal cells, the DNA is then being quantified by using the Nanodrop
