Week 6 – Protocol 1. Prepare a carbohydrate standard curve. a. Add water and a stock solution of D-glucose (2.0 mg/mL) to 6 cuvettes. The volumes are in microliters. Cuvette Blank 1 2 3 4 5 Distilled water 500 400 300 200 100 0 D-glucose solution 0 100 200 300 400 500 b. When all of the cuvettes are set up, add 2.5mL of cold anthrone reagent to each tube. The total volume is going to be 3mL in each cuvette. c. Cover the tops of the cuvettes with aluminum foil. Wrap it around the cuvettes to keep it in place. d. Use the vortex mixer set on 4-5 to mix the cuvettes for 1-2 seconds. This is an exergonic reaction and the contents of the cuvettes will get hot. The layers you saw before in step b should disappear. e. Heat all of the cuvettes at in a water bath at 95 o C for 10 minutes. f. Transfer the cuvettes to the dishpan (with 3 inches of cold water). g. Allow the cuvettes to stand in the water for about 10 minutes. When they have cooled, remove them from the water and blot them dry so water on the
