Genetics 380 : Spring 2009 Name _____________________________ Examination #2A ID # __________________________ March 31, 2009 Section (TA) ___________________ Last year, topics were covered in a different order using another text book This exam has questions from exam 1 and 2 from 2009. As result, it does not add up to 100 points. 1. (10 points) Proflavin-induced wild-type revertants of a number of spontaneous mutants in e + gene of phage T4 were isolated. The amino acid sequence of the affected protein was analyzed in several of these revertants. The affected stretch of amino acids is shown for one such pseudo-wild type revertant (also shown are the amino acids in the true wild type protein). Reconstruct the two mutational events [the original e + -> e - spontaneous mutation (the amino acid sequence of the mutant intermediate is unknown) and the proflavin-induced e - -> e + reversion mutation creating a pseudo-wild type with the amino acid sequence given in the bottom line]. True Wild type (e + ) MET GLY GLU THR LYS ALA GLN .... AUG GGU GAA ACU AAA GCU CAA
