Site of action is monoamine DAT (Dopamine Transporter) ii. Dopamine receptors especially concentrated in striatum iii. Cocaine blocks DAT 1. Blocks reuptake of dopamine, prolonging DA effect 2. 2-3x fold increase in the striatal dopamine 3. Mice lacking DAT are show constant hyperactivity, as if they are always on cocaine (because DA remains in synaptic cleft, no reuptake) 4. If significant loss of DAT (>80%), patients will develop
