Problem Set #5 BIMM100 Winter 2010 – Short Answers 1. Why are ddNTPs required for automated/cycle sequencing reactions but not for pyrosequencing?  They are missing 3’ OH and has a fluorescently labeled base so can look for the sequence of template DNA under a an xray frame of an electrophoresis gel & pyrosequencing doesn’t use ddNTPs because pyrophosphate release gives off light, which is used to find the nucleotide sequence 2. In automated sequencing, which are fluorescently labeled, dNTPs or ddNTPs? ddNTPs 3. List the reagents/enzymes required for automated sequencing. dNTPs, ddNTPs, primer, DNA polymerase, single stranded DNA template 4. Of the two sequencing techniques discussed, which would be preferable to use if  you wanted to sequence an entire genome? Pyrosequencing = whole bunch of genomes generated simultaneously and massively. 5. In pyroseqeuncing, why would it be a bad idea to add all of the dNTPs at the same time?
