MCB 104: Week 2 Practice Problems ANSWERS Section 103/112 1. You are trying to look two membranes separated by less than 100 nm under a light microscope. This is causing you such difficulty. Why is this the case? What other technique could you use? The resolution of a light microscope is its ability to distinguish between two very closely positioned objects. The limit of resolution of a light microscope is 200nm. Therefore, the light microscope is unable to resolve these 2 membranes. You could use electron microscopy, which provides a much higher resolution than light microscopes. 2. You are interested in studying the LAS protein in mermaid cells. First, you wish to determine where in the cell this protein localizes. You have ordered live mermaid cells on the internet and are maintaining them in culture. You have the choice of attaching GFP to your protein of interest, or using an antibody directed to this protein. What do you choose? GFP.
