Unformatted text preview: Lecture 7 Gas Chromatography Mass Spectrometry (MS) Experiment 7 Concept is based on: ◦ Mobile Phase: Liquid or gas that carries compounds through the solid phase ◦ Solid Phase: A stationary composite through which the compound and the mobile phase interact TLC, GC, and LC are chromatographic methods we will discuss GC is actually Gas-Liquid Chromatography Stationary phase ◦ Made up of a liquid polymer coated in column ◦ Inside of the column is open Whole column is heated so sample injected is vaporized A carrier gas (helium) carries the gas through the column ◦ The compounds partition themselves between gas phase and liquid phase many times (theoretical plates) ◦ Compounds separate based on affinity for stationary phase, temperature, and flow rate. Types of columns ◦ Capillary column Wall coated open tubular (WCOT) Liquid phase coats inner tube Support-coated open tubular (SCOT) Solid support coated with liquid ◦ Packed columns Diatomaceous earth (porous, high surface area) Major component is silica Efficiency increases with decreasing particle size Lower particle size, higher pressure needed to move compounds through Types of liquid stationary phases ◦ To get best separation, liquid stationary phase must...
