Wed, March 4 Lecture Transgenic techniques like transformation for animals + plants  For Yeast and bacteria =  transformation of recombinant DNA  DNA is injected into reproductive organs w/ genetic marker. Transgenic organisms show genetic  marker Drosophilia germ-line transformation : (Used for transfrmtn rescue=correct mutant genotypes  w/ recombinant DNA  o P- element   flanked by  2 inverted segmts. These flanking seq’s = recognized by  transposases  (ENZYME!) – cut and paste P element into genome o Efficient transformation of progeny (10-20%)  decreases w/ increasing size of P elemt Mammal germ line transformation (2 ways) 1. Inject vector (RNA virus) containing desired DNA into  nucleus of fertilized eggs   RNA virus  uses reverse transcriptase to convert RNA into DNA   this DNA incorporated into genome of  infected cells a. QUES) If RNA virus uses reverse transcriptase to convert RNA  DNA, then how is it  possible to convert the DNA insert into DNA ??? 2. Embryonic stem cell transformation a. Isolate embryonic stem cells from embryos b. * Transform with recombinant DNA vectors   original embryonic stem cells b/cm  genetically altered stem cells.  c. Incorporate genetically altered stem cells into developing embryo d. Implant embryo
