The rearranged heavy chain gene has the VDJ 5’ to C μ. Prior to antigen stimulation, naïve B cells express IgM and IgD on their cell surface. Both contain the amino acid sequence encoded by the rearranged VDJ.

The rearrangement of Ig genes can be seen using a Southern blot of DNA derived from a cell source other than B cells and comparing it to DNA taken from a transformed B cell line.

The membrane form of immunoglobulin only has a few amino acids extending into the cytoplasm. Thus, in order to signal the cell that antigen has been recognized additional transmembrane proteins are associated with mIg. Ig α and Ig β contain cytoplasmic tyrosine that can be phosphorylated to begin the process of signal transduction to the nucleus which will result in B cell proliferation and differentiation of the cell into plasma cells.
