LAB 11: DNA Isolation Protocol BIOLOGY 1012 - Spring 2010 r WEAR GOGGLES AND GLOVES THROUGHOUT THE ENTIRE EXPERIMENT! r Follow all instructions for waste disposal carefully! (See below.) r Use clean pipettes for each solution to avoid contamination! 1. Label 3 test tubes: A , B , C (and with your initials on each). 2. Place 2.5 ml of E. coli stock suspension (4.0 g /120 ml saline EDTA) into each tube. 3. Add 1.0 ml of 10% (w/v) SDS to each tube and cap tightly. 4. Mix by rotating tubes (gently - to avoid bubbling) for 5 minutes. Suspension will become slightly viscous as bacteria are lysed. 5. Place tubes in a 60°C water bath for 30 minutes. 6. Remove tubes from water bath and cool to room temperature. 7. For each test tube, continue as follows: TUBE A - 1. Follow directions below for spooling DNA. Record results. TUBE B - 1. Place TUBE B into a 100°C dry block for 20 minutes. 2. Remove from dry block & place in test tube rack. Do not cool to room temp. 3. Follow directions below for spooling DNA.
