Insulin Binding: 1. Tyrosine Kinase Receptor 2. outside, inside the plasma membrane going from outside to inside. 3. This receptor activates after insulin binds by autophosphorylating the two beta subunits. There are 3 Tyr residues on the beta subunits. 4. 3 Tyr resides on each of the beta subunits. 5. It activates the target protein by phosphorylating the Tyr residue on the target protein. The phosphate group that is added onto the Tyr residue comes from the ATP that comes in. 6. IRS-1 7. The target protein gets phosphorylated on three Tyr amino acid residues. Insulin Pathway that deals with cell division: 1. Adaptor protein 2. SH2, SH3 3. polyproline 4. PI-3K 5. Grb2 6. GTPase, switches. 7. GEF, GEF proteins turn gtpase on in this pathway by making the gdp leave the sos and allowing gtp to replace it. GAP turns gtpase protein off by allowing it to hydrolyze gtp to gdp. 8. Ras. This protein is activated by GEF and inactivated by GAP. Sos is the GEF protein that helps it activate. 9.
