PHOTOSYNTHETIC CO 2 FIXATION How is CO 2 Used to Make Organic Molecules Such as Glucose? (See Chap 21.7) In 1945, Melvin Calvin & coworkers (UCBerkeley), using 14 CO 2 to investigate photosynthetic CO 2 fixation, found that the earliest labeled product was 3-phosphoglycerate . Further studies revealed that, in reality, that CO 2 was added to a 5-carbon sugar (pentose), to form a 6-carbon intermediate, which then decomposed to form 2 molecules of 3-phosphoglycerate. In other words, the key reaction is CO 2 + C 5 acceptor -> C 6 intermediate -> 2(3-phosphoglycerate) Later it was found that the C5 acceptor is ribulose 1,5-bisphosphate and the chloroplast stromal enzyme catalyzing this key reaction is ribulose bisphosphate carboxylase/oxygenase ( abbr. rubisco), which accounts for 15% of total protein in chloroplast.
