Chronology of Membrane Studies Is there a true physical boundary constraining animal cells? If so, what is it’s composition? Hewson (1775): Human RBCs in normal serum = bi-concave disk in dilute serum = sphere Overton (1890): Tested range of polar and non-polar solutes for ease of diffusion into cells. Observed preferential uptake of lipophilic solutes. Gorter & Grendel (1925): Total lipid extraction of human RBC and measure surface area of lipid monolayer when spread on water. Area = 2 times greater than calculated surface area of RBC. Concept of lipid bilayer . Davson & Danielli (1935): Proposed presence of proteins in (channels)

Unformatted text preview: and on lipid bilayer. 1960s: Electron microscopy extensively applied to cell ultrastucture. 1960s: Alec Bangham Father of the liposome. Karp, Fig. 4.9 Singer & Nicholson (1970): Conceptual breakthrough. Proposed fluid mosaic model of membrane structure. Membrane is dynamic, with lipids and other macromolecules more or less free to move laterally within the plane of the lipid bilayer. Transient interactions are possible. 1980s: Concept of lipid rafts defined as microdomains of unique lipids & proteins in outer leaflet of plasma membrane. Karp, Fig. 4.23 (AFM)...
