Teus 11.11.2008 Page 1 How are the hydrolysis transported to the Ly ????? Here the transport is from the trans Golgi to Ly - the striking thing is the hydrolysis which should transported to the Ly have to receive a remodeling of the glycosyl side chain which are transferred within the ER - so the address change and this done by certain transferase - and this change of oligosaccharide side chain on the hydrolyze is M6P - and M6P recognize by the cargo rece - and this cargo rece with essential coating ptn clathring for the budding of the vesicle - and this transported to the Ly - and the rece retrieve to the Golgi so the rece is recycle - and for this diff kind of coat ptn is used retromer coat which is needed to build up again this vesicle - the release is done by the low PH. The low PH after fusion lead to conf change that the affinity of the hydrolyze to there rece dec 1

Page 2 which structure address the hydrolyze ???? - the striking thing, the hydrolyze it self form a signal patch which recognize by certain domain of the phosphotransferase “PT” - and this PT transfer UDP-sugar residue on the second enzymatic domain …. - This trimming process is one Eg just to show the processing going on within the Golgi to make a certain address for certain ptn If a patient lacking this PT Some of the hydrolysis transfer to diff compartment or even secreted out of the cell And the Lysosomal storage disease it called I-cell disease Lead to huge amount of symptoms it is a fatal disease and it transferred from the parent in a recessive way This mean both partner getting a baby have to have mutated version of this gene and this a homozygous baby mean both partner have transferred the defect on the chromosome 2

Page 3 – 4 Know we are going to learn the transport by the endocytosis - this is impo for some disease - and for understanding the regulation of some hormone processes o to regulate the blood glu in our blood we start with - not every partical has been recognize by the phagocyte is ingested - other cell have don’t eat me signal on the surface it is a broud mechanism to clean up the organs from the bacteria Pinocytosis : vesicle import into the cell And the endocytosis “end” start always with v formation and clathrin coated pit and this again the task of the cell to produce certain patch in the mme by this coat ptn which is come again on the mem and form this v which can enter the cell large no of the mem invaginated by this process this is part of the turn over of the plasma mem 3
