Name:  UB ID #: Date:  BI 130 Lab Assignment #3  Bacterial Operons: Gene Expression and Regulation Due date:  This assignment is to be completed and handed in no later than 11:20  on Thursday, June 24, 2010. Expand the space below each question and insert your typed answer and then  submit a printed hard copy by the due date. The  lac  operon This assignment focuses on the regulation of gene expression in bacteria. The control  over the synthesis of gene products (proteins) often occurs at the level of transcription  of the gene and is mediated by protein -binding regulatory sequences associated with  genes . Once bound, such proteins exert their regulatory effect by either activating or  repressing the gene’s transcription . This was first realized through work attributed to  Jacob and Monod (done in the 1940’s for  lac  and 1950’s for  trp ), who described  regulatory systems in  E. coli  – the  lac  operon and the  trp  operon – which functions in  turning certain genes  involved in the metabolism of  lactose  on and off for  lac  or turning  certain genes  involved in the metabolism of synthesis of tryptophan on and off for  trp depending on the nutritional environment of the cell.  Innumerable variations on the  basic processes exemplified by the  lac  and  trp  operons have been (and continue to be)  discovered in both bacteria and eukaryotic cells.
