Student#504310 Yeast fermentation lab Purpose To determine if CO 2 output is affected by temperature and addition of additional sugars. Methods 2 mL of yeast, 2 mL of distilled H 2 O, and 2 mL of a 20% glucose solution will be added to 9 separate test tubes. In addition, 3 test tubes will contain 2 mL of a 20% fructose solution, 3 test tubes will contain 2 mL of a 20% sucrose solution, and there will be 3 test tubes that will act as a control having just an additional 2 mL of H 2 O added. For the first experiment, prior to the addition of the yeast, 1 tube of the control, 1 of the fructose, and 1 of the sucrose will be added to a 50˚C water bath and allowed to sit for 5 minutes to stabilize the solution’s temperature. Then the yeast will be added and the tubes will sit for 5 additional minutes. The yeast/sugar solution will then be drawn up into a respirometer and the measurements will be recorded every 2 minutes until 20 minutes has been reached. After the 20 minutes has been reached, the CO 2 levels in the respirometer will be recorded every 5 minutes until 40 minutes has elapsed at which time the experiment will be concluded. For the second experiment, prior to the addition of the yeast, 1 tube of the control, 1 of the fructose, and 1 of the sucrose will be mixed at room temperature (25˚C) and allowed to sit for 5 minutes to stabilize the solutions temperature.
