Aseptic Technique - 1. Hand washing before and after the experiment 2. Disinfect bench area before and after the experiment 3. Sterilize the inoculating tools before, after and during the experiment 4. Never leave inoculating tools on the bench top 5. Dispose of any contaminated material in the biohazard bag 6. Never put bacterial culture down the sink Contamination – Disinfectants – *Use 10% Clorox and Lysol as disinfectants Sterilization - How can sterilization be accomplished? 1. By heat – using a Bacti-Cinerator to sterilize the loop and needle 2. By steam under pressure – using an autoclave at 121 degrees C and 15 psi for 15 minutes for large scale sterilization. Innoculation – the process of introducing a MO into a fresh, sterile medium in order to initiate the growth Incubator – after inoculating the medium with MO, the culture must be incubated at 35-37 degrees C for 24-48 hours for optimal MO growth. *Invert the agar plate when placing it in the incubator, so the condensation collected inside the cover of the agar plate will not drip to the surface of bacteria. Microscope – Compound Binocular Light Microscope *Know the different parts diagram pg 6 What is the condenser lens used for? to focus the light from the lamp through the specimen to be viewed. What is the iris diaphragm used for? to control the amount of light that comes through
