September 22, 2010 DNA Replication Parental strand can be used as a template for the formation of a new daughter strand that is complementary to the parental strand Base pair sequences are complementary Strands are anti-parallel Matthew Meselson and Franklin Stahl o Base-pairing template model could proceed via a conservative or semiconservative mechanism Conservative o Two daughter strands form new double-stranded DNA molecule. Parental duplex remains intact Semiconservative o Parental strands are separated and each acts the template for the formation of a new daughter strand Meselson and Stahl experiment o Nitrogen exists as N-15 or N-14 o Ammonium salts enriched with N-15 o Grew E. coli in an environment rich in N-15 then switched them to an environment rich in N-14 o They extracted and sampled the DNA o Started with DNA where all cells have N-15 (heavier isotope) o After the switch in medium occurs they sampled the DNA to determine the amount containing each isotope
