September 7, 2010 Review from Last Class Have to remove paraffin for Light Microscopy because you need to stain the sample (H and E) which are aqueous solutions so in order to let the stain hold you need to remove the non-aqueous paraffin Lipids and fats dissolve in ethanol Electron microscopy o Electron dense structure is dark Desmosones are like porcupines (dark, spines on electron microscopy pictures) Epithelial Cells Part I Functions of the Skin o Protection-Resists abrasion and penetration and blocks the entry of foreign material o Thermoregulation – Skin controls the transer of heat across the body’s surface, facilitating as needed both heat retention and heat dissipation o Sensory o Communication – skin releases visual and pheromonal signals o Immunological defense o Appendages – hair, nails, sebaceous glands, sweat glands Hypodermis is packed with fat cells that have dissolved during the fixation and dying process Epidermis and dermis are separated by the basement membrane o Will not see basement membrane in the lab except in hair follicles or the respiratory tract o Special stains bind the basement membrane to make it pink, cannot see
