(4) Chromatography Some HINTS for chromatography HINTS: 1. When setting up the Pasteur pipet column, make sure that you use a very small amount of the ferrocene mixture. General rule of thumb for newbies is that if you think you have enough, use half that amount. 2. MAKING SPOTTING SOLUTIONS FOR TLC. use a very small amount of crystals for spotting the TLC plate. General rule of thumb is that you would use five small crystals for three to five drops of methylene chloride. Seriously, that is all you need. What usually happens is that students spot way too much compound and the TLC needs to be re-done. That really, really wastes your time in the lab. Even though I mentioned it several times in the lab, it seems like either students find it incredulous that such a small amount could be used, or that they think they would rather find out the long way. 3.
