Types of blood vessel development: 1. Vasculogenesis 2. Angigenesis: a) Non-sprouting angiogenesis (by intussusception or by bridging); b) Sprouting angiogenesis. 3. Angiogenesis is the process of new blood vessel formation from pre-existing vessels, and is important in embryonic development and wound healing. 4. Endothelial cells form monolayers, with cells adhering to each other via VE cadherin and PECAM, and interacting to the underlying basement membrane (extracellular matrix) via integrins. 5. Angiogenesis process: angiogenic factors bind their receptors on endothelial cells, leading to: a. Cell-cell de-adhesion through phosphorylation of the cell adhesion molecules b. Loss of interaction of cells to the matrix through production of proteases that degrade the matrix c. Increased cell proliferation and migration up the angiogenic factor gradient to form a sprout d. Fusion of two sprouts to form a continuous vessel e. Recruitment of periendothelial cells to stabilize the vessel 6. Angiogenesis can be associated with pathological events, including tumor-induced angiogenesis leading to tumor
