Light grown barley leaves and dark grown barley leaves are grinded with methanol and the extract is used for spotting . Spotting was done on the plastic backing sheet. After spotting every time, it was dried with a air hose, and spotted 15 times until there was a clear, dark spot. Then, the sheet was put it in ethyl acetate, and we observed the spot travel by capillary action. The rest of the sample was analyzed in the spectrophotometer.
