Santiago Rubio TMS Lab Section 206 Dr. W. Krause February 19, 2010 Experiment 3: Micrometry and Microscope Areal Analysis 1. These were the following steps used to calibrate the eyepiece micrometer. The stage micrometer was placed on the microscope stage and focused at 100x magnification. Using the stage dials and controls, the first division line of the eyepiece with the “0” mark was aligned with the first division line of the stage micrometer. At 100x, 50 stage units were equivalent to 50 eyepiece units. Each mark on the stage unit was 10µm apart. This made each eyepiece “unit” 10µm. The stage micrometer was then focused at 400x magnification and adjusted to align the eyepiece lines with the stage lines. At 400x, 10 stage units were equivalent to 40 eyepiece units. Since each stage unit was 10µm apart the following equation was used to calculate the conversion factor. 10 stage units x 10µm/stage unit
