Chapter 7 Cell Test Tasks 1. Magnification is the ratio of an object’s image to its real size, while resolving power is a measure of the clarity of the image; it is the minimum distance two points can be separated and still be distinguished as two separate points. 2. Light microscopes pass visible light through the specimen and then through the glass lenses which refract the light to magnify the image. At greater magnifications with light microscopes the image becomes blurry. Some advantages are its availability and ease of use. The Electron Microscope focuses a beam of electrons through the specimen or onto its surface. Transmission electron microscopes focus through the specimen and are great for inner views but require extremely thin specimens. Scanning electron microscopes scan the surface of the specimen and give great 3-D images and detailed pictures of the surface, but don’t show the inner workings as well. 3. Fractionation begins with the homogenization, or disruption of cells. After spinning it in a centrifuge on low speed the pellet and supernatant will separate and the supernatant can be decanted into another test tube and centrifuged again at a higher speed, and this process is repeated. The point is to separate the cells organelles without damaging them so they can be viewed separately. 4. Prokaryotic cells lack a true nucleus and other membrane-enclosed organelles of the eukaryotic cell. 5. There are upper and lower limits to cell size because the lower limits are set by the smallest size that has enough DNA to program and carry out metabolism.
