FA may enter into other cells by diffusion across plasma membrane, but likely facilitated by other transport proteins (more research is required). FA in cytoplasma is escorted by FABP to MT FA cannot get into the mitochondria by diffusion!! FA breakdown mainly occurs in mitochondria matrix 4 Activation of Fatty Acids Fatty acids are activated upon entering cytoplasm of target cell. Reaction: Fatty Acid + ATP + CoA <=> Acyl-CoA + AMP + PPi Enzyme: Acyl-CoA synthetase Several forms specific for different chain lengths. Associated with mitochondrial outer membrane. Requires Mg ++ Reaction driven by hydrolysis of PPi, overall G o = -34 kJ/mole. 5 Acyl-CoA synthetase mechanism: Two steps: 1. Formation of Acyl-adenylate intermediate. 2.
