Use citrate as carrier molecule. Uses ATP to drive synthesis of thioester Provides cytoplasmic NADPH for reductive FA syn Figure 20-24 Acetyl-CoA in MT is transported into cytosol as citrate 5 Acetyl-CoA is activated by adding a CO 2 , generating Malonyl-CoA, the donor for fatty acid synthesis. Acetyl-CoA carboxylase (biotin) (ACC): Acetyl-CoA + HCO 3- + ATP --> Malonyl-CoA + ADP + Pi Two step reaction, similar to pyruvate carboxylase and propionyl-CoA carboxylase. Page 703 First committed step; subject to allosteric and hormonal regulation Citrate stimulating; LCFA inhibiting feedback inhibitors finer regulation by kinases on 6 sites: Ser76- P inactivates ACC insulin stimulates de-P of Ser76, activates ACC 6 FA synthesis 7 enzymatic steps E. coli: independent individual enzymes makes FA as precursors for phospholipids membrane...
