Four types of psychostimulants: % amphetamine % cocaine % caffeine % nicotine 12/3/2008 2 % Amphetamine % Increase in alertness and decreased sleep Amphetamines • Amphetamine and related psychostimulants (e.g., methamphetamine) resemble the neurotransmitter DA in their chemical structure. % Increase in alertness and decreased sleep % Increased focus (sustained attention tasks) % Reduced appetite % Positive affect (mood) % Low doses very effective % High doses interfere with performance and concentration % Sid ff t f h t i % Side effects of amphetamines: % dysphoria % Hypersomnia (2 ‐ 3 days) % Stereotypy (teeth grinding) % Restlessness % Paranoia % Inflated self ‐ importance % Narcolepsy % ADHD (Attention Deficit/Hyperactivity Disorder) % Weigh loss Clinical Use of Amphetamines Mechanisms of Amphetamine Action % AMPH and METH are indirect agonists % cause DA molecules to be release into cytoplasm of nerve terminal % these DA molecules are then transported outside of the terminal by a reversal of DAT % resulting in massive increase in synaptic DA concentrations 12/3/2008 3 Mechanisms of Amphetamine Action...
