What are the roles of : A) anion hole and; B) serine in a serine protease? Anion hole – It stabilizes the tetrahedral intermediate and the transition states for the formation and breakdown of this intermediate. Serine – It attacks the amide bond (carbonyl carbon) and forms the acyl enzyme intermediate. 2. What are the two key tenets of microscopic reversibility? 1 – All reactions are reversible and 2 – the pathway followed in the forward direction will be followed in the reverse direction (or, a catalyst will catalyze both the forward and reverse reaction). Now, answer any 14 of the next 16 questions. 3. Briefly outline the process by which a new drug is developed. 1. Choose a disease and enzyme target. 2. Develop an assay and screen compounds. 3. Synthesize analogues of lead compounds to find better inhibitors. 4. Test compounds in cells. 5. Test compounds in animals 6. Develop good synthesis of compound. 7. Phase I-III testing in humans – look for efficacy and avoid side effects. 4. Suppose that life tried to evolve using by attaching fatty acids to ethylene glyol, rather than to glycerol.
