Ilana Fuzaylova Lab #1 Introduction In this lab we will be able to monitor catalase activity on H 2 O 2 , boiled H 2 O 2 and on live tissue. We will see the effect of extreme temperature on enzyme activity. We will also be able to test and observe the results of a timed catalase reaction and the reaction rates for catalase decomposition of H 2 O 2. Materials: Chemicals 1.5% H 2 O 2 Catalase solution in medicine cup Beaker, 250 mL with water for boiling catalase solution Container with ice for catalase solution 70 mL 1.5% H 2 O 2 in 120 mL graduated cup distilled water in medicine cup catalase solution in medicine cup 1.0 M H 2 SO 4 in glass beaker 2% KMnO 4 in 120 mL graduated cup Specimens 1 cm 2 cube of liver and/or potato Instruments Apron Gloves Goggles 3 medicine cups test tube test tube holder wood macerating stick 12 mL syringe mL pipet Marking pen or pencil Labeling tape Hot plate Knife or scalper for cutting cubes of potato and/or liver medicine cups 6-mL syringe plastic marking pen or pencil, labeling tape, stopwatch, paper, white Procedure: The Uncatalyzed Rate of Hydrogen Peroxide Decomposition Remove 10mL of the H 2 O 2 from the cup and transfer it to another medicine cup labeled “Uncatalyzed H 2 O 2 Activity.”

Add 1mL of distilled water to the H 2 O 2 in the Uncatalyzed H 2 O 2 Activity cup. Add 10mL of 1.0M H 2 SO 4 to the cup. Label the remaining medicine cup “Uncatalyzed Assay.” Transfer 5mL of the Uncatalyzed H 2 O 2 Activity mixture to this cup. Draw 10mL of 2% KMnO 4 into the 12mL syringe. Read the initial volume of KMnO 4 in the syringe and record this in Data Table 1. Holding the 12mL syringe over the cup, carefully depress the plunger so that one drop of KMnO4 falls into the cup. Swirl the cup gently and observe the solution for any pink to brown coloration that remains after mixing is complete. Complete the titration by repeating the step before, drop by drop, until a pink to brown coloration of the solution persists after mixing. Record the final volume of the KMnO 4 solution in the 12mL syringe in Data Table 1. Test of Catalase Activity Part A : Observation of Catalase Action Place 10mL of 1.5% H 2 O 2 into a medicine cup labeled “Catalase Extract Activity.” Add 1mL catalase solution to the H 2 O 2 in the medicine cup. Gently swirl the cup to mix the contents, and observe the reaction occurring in the cup. Record your observations in Data Table 2. Part B
