Within 50 bp of the primer the data are unreliable due to inaccurate migration of short DNA sequences with fluorescent dyes. In large fragments, the peaks spread out due to diffusion. Becomes difficult to resolve peaks when relative mass differences are small. Signal intensity decreases as fragment length
