In order to understand Statin function, you will have to characterize its transcriptional regulation. As a first step in this process, you decide to isolate the promoter of the Statin gene. a. Describe a strategy for this. b. Once you have isolated the putative Statin promoter, how can you test its activity? Be sure to describe the reagents you need for these studies. c. Describe a strategy for identifying sequences in the Statin promoter that are important for transcription activation. What will your data look like if there are four regions of the Statin promoter that contribute to transcription activation in human epithelial cells. d. You isolate a 185 base pair DNA sequence that is necessary and sufficient to promote Statin transcription. Now you want to identify proteins that bind the promoter.
