Lab 9 outline; extraction of DNA from bacterial cells ( E. Coli ). Read lab 9 in the lab manual. Your protocol should include the information needed to perform the following steps to successfully obtain DNA. Check the manual for details A. breaking open the cells – Lysozyme (enzyme to digest cell wall) B. open the plasma membrane – SDS (fancy detergent, think Tide) C. prevent destruction of DNA by nucleases: EDTA (what are nucleases?) D. inactivate DNases by heating (inactivate enzymes w/o destroying DNA) E. remove proteins from DNA with NaClO4 and chloroform-isoamyl alcohol F. precipitate DNA and remove water with ethanol G. dissolve and store DNA in STE salt – maintain integrity of double helix You should know why salt is necessary from Dr. Rappaport’s lectures The DNA will be stored in the STE until the next lab for analysis There is an interesting web link in the lab folder and on Dr. Waring’s web page.
