Extraction Lab Protocol Prepare by organizing the lab bench with enough 1, 5 & 10 ml pipets, the proper size pipet pumps, the tubes, parafilm. Find out where each solution is that you will need. Work as a pair. Synchronize your steps with the others in the lab. Label prep with group initials to identify all tubes. 1. Pipet 4.5 ml of bacterial suspension into 15 ml screw cap tube (color?) 2. Pipet 0.25 ml of lysozyme solution into the sample and mix by gentle rolling/inversion. Incubate prep in 37C water bath for 20 minutes . (invert about every 5 minutes in bath) 3. Pipet 0.5 ml of 10% SDS and mix by gentle rolling/inversion for several minutes. Look for the solution to turn viscous as you slowly invert the tube and become clear. 4. Incubate the prep for 10 minutes in a 60C water bath. 5. Let the prep come back to room temperature for about 10 minutes. 6. Add 1.25ml of NaCLO 4 and then 6.5 ml of chloroform/isoamyl alcohol. Secure the screw cap tube with parafilm and mix by hand for 10 minutes
