Properties of DNA polymerase: 1. Needs to have a template. 2. It can only add nucleotides to the 3’ hydroxyl (OH) of a nucleotide. Therefore DNA replication always proceeds in the 5’ to 3’ direction. 3. DNA polymerase can only add nucleotides to to an existing single strand of nucleic acid. Therefore DNA synthesis must always be primed by a small piece of RNA or DNA. For replication of chromosomes RNA primers are used. For the polymerase chain reaction (PCR), DNA primers are used.
