RECOMBINATION COMPLEMENTATION Tests for: gene location gene function Infect: B (permissive): K (non-permissive): Why: Allows recomb to happen not growing, so recomb does NOT happen! How does this work? Both rIIA and rIIB proteins required for infection to proceed. Without one, infection is aborted. If complementation occurs, infection can proceed and the phage progeny can lyse the cell. These phage have the mutant “parental” plasmids! Because NO recombination has happened! They would never be able to lyse alone, they can only do it when theyre both present Plate: K (tells you how many recombinant) B (because the progeny Why: B (tells you how many phage total) phage is rII-!!) “+” means: mutations NOT in same nucleotide mutations NOT in same gene “-“ means: mutations DO overlap mutations ARE in same gene if “+”, expect: B > K B = K if “-”, expect: B >> K B >> K b.
