Captive breeding programs at zoos are struggling due to the sterility of some mating pairs. In each case of sterility, the male produces no motile sperm. The zoo veterinarian suspects that one of three LINKED genes may be to blame. She develops PCR assays for the polymorphic markers A, B, and C (microsatellite loci) that are tightly linked to each gene. She uses these assays to analyze DNA from male and female cheetahs at the San Diego, Washington DC, and St. Louis zoos. A gel with her results is shown to the right. (a) Why do the males have three bands? These three linked loci are on the X chromosome. Males have only one X and no alleles on the Y. (b) Why do the females have five bands? Although females have two X chromosomes and could have as many as 6 bands, each is homozygous for one of the markers, resulting in only five bands. (c) Based on these data, which allele is associated with the sterility? B1 Over the years, the Washington DC pair produces many offspring. PCR analysis of the DNA from 8 sons reveals the inheritance pattern shown to the right. (d) In the mother, which A, B and C alleles are present on each homologue?
