Lecture 5 I. Two faces of telomerase A. End replication problem PIC ! -telomerase-extends 3’ ends of telomerases and extends template of lagging strand at the 3’ end and was discrovered by Blackburn and Greider. Components of telomerase: -RNA-template – repeadted sequence ~6nt per binding -TERT (telomere reverse transcriptase) – copies RNA to DNA. Uses RNA to syn DNA -telomerase will base pair, binds to RNA template of telomerase, add additional six neucleotide. … ???? add B. Bridge-breakage fusion -telomere looks like double stranged break so cell want to repair this. Telomeres can protect the ends of chromosomes form shortening. PIC 2 (look at wiki) -telomerase is normally expressed in dividing cell such as embryonic or stem cells or cancer cells -after crisis stage, telomerase gets activated. Telomerase is almost always activated in the cancer cells by mutation. -3’ end can form a loop and invatde into the strand. Makes a d loop and invade C. Telomere Protection; t-loop PIC 3 D. Hayflick limit; elixir of life?
