Karlo Natonton Biology 140-56 An Investigation into the Action of Enzymes Based on our findings, we can conclude that with an increase in substrate concentration comes an increase in enzyme activity. We can also conclude that as the temperature that the reaction occurs in increases, the enzyme activity of the reaction increases as well. The pH experiment resulted in enzyme activity being highest at pH 4. Experiment A: Effect of Substrate Concentration on Enzyme Activity In Experiment A, we tested the effect of substrate concentration on enzyme activity. We tested 5 different cuvettes with each separately containing 0mM, 1mM, 5mM, 10mM, and 15mM of ONPG. After observing the reactions for each cuvette, we discovered that as the ONPG concentration increased, so did the enzyme activity.
