2 1. The figure below shows a single nucleotide polymorphism ( SNP ) between two alleles that creates a BamHI restriction fragment length polymorphism (RFLP) in a 10 kb stretch of chromosomal DNA. The RFLP can be detected by Southern blot analysis using a radiolabeled probe complementary to this region. Because this SNP is associated with the Sickle Cell Anemia, individuals can be screened for this autosomal recessive disease. Three individuals submitted their DNA for genetic testing to determine their risks for the disease. The Southern blot analysis reveals the following bands. Individual 1 1.5kb Individual 2 1.5kb and 4kb Individual 3 4kb (a) Which individual will definitely get the disease? (5 pts.) (b) Which individual is a carrier of the disease allele? (5 pts.) (c ) Which individual is not a carrier of the disease allele? (5 pts.) (d) If a probe corresponding to the 8kb region were used instead of the one corresponding to the 6.5 kb region, what new banding pattern would be indicative of a carrier of the disease. Circle the correct answer(s) below. (5 pts.)
